When creating a Button object in the interface, you can change the image. Changing to a transparent-background image will still cause the button to have a (by default) gray background. When changing the button's type from Pushbutton/Pushbox to Checkbox/Radio and then back to Pushbutton/Pushbox the transparency is then applied. However, when closing the interface and re-opening, the transparency is lost.
The desired behavior is to have the transparency as an additional check-box to include transparency or not. Then, the desired behavior is to apply it and not lose it's function when closing/opening the interface again.

What control is beneath the button?

A map.

Transparent anything doesn't work over the map. Although ideally it'd be good to have Dream Maker refresh this properly, ultimately you'll be disappointed in Dream Seeker's performance on this anyway.

Honestly I wasn't even looking at utilizing it. A forum post related to this was regarding using buttons instead of labels because of background-images for labels are easier to accomplish due to transparency compared to buttons, but labels don't offer click-ability. When experimenting on this, I noticed this behavior and thought it odd.

Labels won't work over the map for transparency either (except in the webclient). Transparency is really, really limited in Windows.

Yeah, the members have suggested web-client features such as that. Said topic is here: http://www.byond.com/forum/?post=1883708
