So, I know DMI is a sort of modified PNG format (assuming this because I can open dmi files in a PNG editor), but what's the format for defining icons? The reason I ask is that I need to make a dmi file with ~343 different icons, and I'd much rather just write a script to generate the file, and save myself a _ton_ of work. Is the format documented anywhere for people to use, or is it private and I'll just have to make the file semi-manually?

Also I wasn't sure where this thread should go, but this seems the most appropriate place...
DMI version 2 (or, if you will, DMIs created in BYOND 4.0) is just an ordinary PNG file. All the needed formatting and information is stored as comments/metadata inside the file. You can view the comments with an editor and easily work out the simple format.
There was also this topic about the DMI v2 format.
