Does the problem occur:
Every time
This includes on other hard drives.

Did the problem NOT occur in any earlier versions? If so, what was the last version that worked? (Visit http://www.byond.com/download/build to download old versions for testing.)
I've tried 500 and 506, neither work.

I should mention I can open an existing one perfectly fine. Which gives me somewhat of a workaround but it's still agrivating.
Does the directory desktop/test exist? Personally, I've only seen this when it does not.
No it doesn't however in previous versions of DM I found it created it for me and I found that rather useful.

Regardless, creation isn't working even when the directory does exist.
Have you tried rebooting your system or closing some other apps? I've found the system sometimes gets into a state where it has way too many handles and doesn't know what to do with itself.

And are your other projects in that same desktop dir as well? Did something possibly change with the permissions?
After a reboot (And a reinstall, because whatever) I can successfully make a project within a folder already created but I'm still unable to create a folder directly with the program, was that functionality removed or something?
No, the functionality to create a project folder was not removed. This sounds very much like a permissions problem on your system.
I have full admin access and UAC is turned off, the program is also set to run as admin by default, what more to do?
Is the folder read-only or anything like that? Have you tried making a new environment in a completely different folder?
As mentioned I've tried on all three of my Harddrives and none of them have worked.
Another thing that just occurred to me is that it's possible that antivirus software could interfere with I/O. What antivirus are you using, and what happens if you temporarily disable it?
I don't have an active antivirus running, however Malwarebytes is installed in the background doing whatever it does, I haven't been able to find a way to disable it so I'll just uninstall in a minute.
